Restriction digests of the clone give the following sizes (kb): BamHI--10.0, 1.3, 0.55; BglII--11.0, 0.9; PvuI--6.0, 4.0, 1.5, 0.95; PvuII--3.2, 3.0, 2.4, 1.8, 1.4, 0.45; SacI--12.0. The insert contains the following restriction sites (approximate kb from the 5' end): BamHI--1.21, 1.75; BglII--1.43; PvuI--1.28; PvuII--0.12, 1.86; SacI--1.37. Constructed using NheI adaptors. Most of the insert (all but approximately 250 bp) can be excised using BamHI, but the insert will be in two fragments- 1.2 and 0.55 kb. Contains the complete coding sequence only. |
